The synthesis of cholesterol in the liver through Acetyl CoA is the first step, after several complex changes an intermediate called 7-dehydrocholesterol is reached. When UV rays come into contact with the skin, this 7-dehydrocholeserol undergoes transformations, producing vitamin D3. This vitamin D3 is not biologically active, therefore it must undergo two hydroxylations: the first in the liver forming 25-hydroxycholecalciferol (calcidiol) and the second in the kidney forming 1,25-dihydroxycholecalciferol also called calcitriol (active form).
